Burkholderiapseudomalleiis a soil saprophytic Gram-negative bacteriumwhich causes meloidosis, a life-threatening disease with high mortality. Evenextensive studies have been exploit the treatment of meloidosis, there is stillno licensed vaccinate to prevent infection of the bacterial.B.pseudomalleiis commonly found in soil at a depth of 25-45cm and can move to the surfaceduring the rainy season. The rice farmers of Thailand seldom wear protectivefootwear, so they are very vulnerable to this pathogen throughcutaneousinfection. To find the natural phage against this bacterial seems important toprotect the rice farmers from meloidosis. The researchers collect 101 soilsamples from the endemic regions. They have isolatedB.pseudomalleibacteriaand their associated phages in nature. Most samples only have bacteria orphages, only a few soil samples have both bacteria and phages together. Based ontheir argument, they underestimate the abundance of the bacteria because theyincubate the organisms at 40℃. At this temperature, the lysogenized phage can be induced out, and they will kill all the bacteria, that is the reason why they got several soil samples with only phages in them. They observed the morphology of phages induced out by mitomycin C (MMC) with transmission electron microscopy. The widely isolated and induced phages are all icosagon. They analyzed the genome of the induced and isolated phages and the results suggest that the induced phage is quite similar to the wide type phage but not identical. The wide type and induced phages are all belong to podoviruses.
