        重组耐碱蛋白A在天然金黄色葡萄球菌蛋白A的基础上对蛋白质结构进行了改造,与天然蛋白A相比,去除与Fab的结合位点,增强重组耐碱蛋白A与Fc结合的专一性,同时提高蛋白稳定性和耐碱性。重组耐碱蛋白A与琼脂糖凝胶键合后可制备用于抗体纯化的亲和填料,可从腹水、血清、细胞培养上清或细胞抽提物中分离和纯化多种哺乳动物不同亚型的抗体或包含抗体Fc片段的基因工程重组蛋白。重组耐碱蛋白A的C端加入了一个半胱氨酸以增强其同凝胶的定向共价键合。

Recombinant alkali resistant protein A has modified the protein structure on the basis of natural Staphylococcus aureus protein A. Compared with natural protein A, it removes the binding site with Fab, enhances the specificity of recombinant alkali resistant protein A in binding to Fc, and improves protein stability and alkali resistance. The recombinant alkali resistant protein A can be bonded with agarose gel to prepare an affinity filler for antibody purification, which can separate and purify antibodies of various mammalian subtypes or genetically engineered recombinant egg white containing Fc fragments from ascites, serum, cell culture supernatant or cell extracts. A cysteine was added to the C terminal of recombinant alkali resistant protein A to enhance its directional covalent bonding with gel.
