研究背景
动脉粥样硬化(AS)性心血管疾病作为全球人口死亡的重大疾病之一,其发病常与营养膳食不均密切相关。因此,人们通过摄入可降低AS发病风险的含非营养素成分的果蔬来防控AS的发生、发展。
有研究证实,巨噬细胞炎症是AS发病的重要因素,且受NF-kB(p50、p65亚基)异二聚体调控。在休眠期的巨噬细胞中,胞浆中的NF-kB与其抑制剂IkBα结合,但当巨噬细胞遭受促炎因子刺激时,IkB激酶(IKK)被激活,进而使IkBα磷酸化,导致IkBα发生泛素化及蛋白酶体降解;NF-kB被释放后,NF-kB核定位序列暴露,由胞浆动力蛋白Dynein将NF-kB以负极运动方向运输至胞质蛋白importin-5上,importin-5护送NF-kB从胞质向核内转移,启动多个基因的转录,例如促炎因子IL-1β、TNF-α的表达,那么,抑制巨噬细胞中NF-kB激活可能是治疗AS的潜在途径。倍半萜类化合物山莴苣苦素(Lactucopicrin,LCP)作为一种分布于菊科蔬菜生菜、菊科药用植物菊苣中的苦味物质,具抗疟、抗癌、镇痛镇静等药理活性,但其对AS的影响未见报道。故中山大学公共卫生院王冬亮教授团队基于RAW264.7巨噬细胞模型和ApoE基因敲除小鼠模型探讨LCP对AS的影响及功能机制。
