What is MD?

As everyone knows, cancer is a terrible disease. Chickens also get cancer. Marek’s disease (MD) is a T cell lymphoma disease of domestic chickens induced by the Marek’s disease virus(MDV). In recent decades, the economic impact of MD on the worldwide poultry industry is substantial because of losses caused by mortality, reduced egg production and meat condemnations. Although vaccination greatly reduces MD incidence and symptoms, it does not prevent MDV infection, replication, and horizontal spread. Furthermore, MD still remains a threat because of unpredictable and sporadic outbreaks of highly virulent strains of MDV combined with the incomplete immunity that is elicited by vaccination.

What is microRNA?

MicroRNAs (miRNA) are a new class of noncoding RNAs, which are encoded as short inverted repeats in the genomes of animals and plants. In view of their importance, miRNA genes become a research hotspot.Recently,studies also have shown that miR-181a plays an important role in many cancer diseases .Especially it was found to be related to tumor transformation in chicken MD.

What is ANP32A?

ANP32A is a tumour suppressor whose expression is altered in many cancers and involved in many activities and biochemical processes. Its shape looks like a group of tadpoles.Previous research found that the gene ANP32A was verified as a direct target gene of miR-181a. Moreover,the researchers point out this gene is highly expressed in MD tumor tissues.

What did we do?

In this study ,to elucidate the role of ANP32A gene in MD tumor transformation , RNA interference technology were used to block its expression,and we also analyzed the biological effects of ANP32A knockdown on tumor cells using CCK-8 and transwell .A siRNA with interference efficiency of 70% was transferred into tumor cells.

What did we find ?

The results showed that tumor cell proliferation and cell cycle was unaffected after transfection,but it is notable, cell apoptosis was inhibited remarkably,which indicates that ANP32A gene may be an inhibitor of tumor cells by promoting apoptosis.This research established foundation to reveal the mechanism of MD.
